Abstract: | In this study, our interest has been focused on the in vitro properties of the complexes formed between two peptide Bombesin derivatives and the radionuclide 99mTc. We have studied their metabolism in homogenized liver and kidneys, stability in vitro in human plasma and cancer cells PC-3 and also releasing time of radioactivity from cancer cells. The metabolism is checked by HPLC comparing chromatographic peaks of radiolabeled peptides before and after homogenization procedure while the time increases. Stability in vitro is checked determining labeling efficiency after their incubation in plasma and cancer cells. It is concluded that two Bombesin derivates appeared low stability in homogenized liver and kidneys and high stability in human plasma and cancer cells PC-3. The comparative studies between two derivates (BN 11 and BN 11p) showed that BN-11p appears promising for the in-vitro properties, so it may be a useful tool for early tumor detection. |
Abstrakti: | Peptidet e shenuara me radioizotope 99mTc, zoterojne nje potencial te madh si radiofarmaceutikate. Permes ketyre peptideve, Bombesina dhe Gastrin-Releasing Peptide kryejne funksionin e faktorit rrites autocrine ne disa qeliza neoplastike. Ky studim eshte perqendruar ne vetite in vitro te komplekseve te formuara midis dy derivateve peptidik te Bombesines dhe radionuklidit 99mTc. Studiuam metabolizmin e tyre ne melci dhe veshka te homogjenizuara, stabilitetin in vitro ne plazem njerezore dhe qeliza kanceroze PC3. Nga kombinimi i rezultateve u arrit ne perfundimin se dy derivatet e Bombesines ne studim kane stabilitet te ulet ne veshka dhe melci si dhe stabilitet te larte ne plazem njerezore dhe qeliza kanceroze. Studimet krahasuese midis derivateve BN11 dhe BN11p treguan per permiresim te vetive in vitro te derivatit BN11p, gje qe i jep atij mundesine per t'u perdorur me sukses si radiofarmaceutikati me i pershtatshem ne diagnostikimin e kancerit te hershem ne pankreas. |
